  Dipcadi (Asparagaceae, Scilloideae) has long presented phylogenetic and taxonomic challenges. A recent study by Chapano et al. (in press) based on ITS sequence data found several non-monophyletic species and an unresolved backbone topology of the genus. In this study, we present the first high-throughput sequencing (HTS) phylogeny of Dipcadi, using data from target capture of the angiosperms353 loci. The sampling is focused on African diversity. Additionally, we provide an updated ITS phylogeny representing 15 Dipcadi species, including 18 newly sequenced accessions. We expand taxon sampling, especially of D. longifolium, to further explore phylogenetic relationships and species limits in the genus. Although the angiosperms353 loci did not fully resolve the backbone topology of Dipcadi, both approaches generated similar topologies with six well-supported clades, but with incongruences regarding deep nodes. We corroborate monophyly of D. marlothii and D. glaucum. Dipcadi longifolium represents a species complex, possibly including D. gracillimum, but we refrain from taxonomic revisions based on the current sampling. Dipcadi viride and possibly D. platyphyllum are paraphyletic, warranting further attention. Our findings confirm that ITS can be effectively employed as a barcode marker in Dipcadi.

  The phenological and morphological variation studies among forms of Laggera Sch. Bip. Ex. Benth. and Hook species are limited, despite the medicinal use of the genus. Therefore, this study aimed to document phenology and morphological variation in cultivated populations of Laggera alata and Laggera crispata forms from seedling to maturity. The forms were categorized as Laggera alata with a small capitulum (LA-SC), Laggera alata with a large capitulum (LA-BC), Laggera crispata from South Africa (LC-SA), and Laggera crispata from Zimbabwe (LC-ZIM). Seeds were germinated in Petri dishes, transplanted to plug trays, and later to field plots at 60 days. Phenological events were recorded when observed in at least one plant. Twelve qualitative and four morphometric traits were measured monthly on five plants per Laggera form. Analysis of variance and Tukey’s Honestly Significant Difference test (p < 0.05) were used for data analysis. Results indicated significant variation in phenology, qualitative traits, leaf traits, plant height, and stem diameter both within and between L. crispata and L. alata forms. Morphometric traits, such as leaf size and the number of leaves per plant, were identified as key descriptors for differentiating L. alata forms. These findings provide a foundation for the introduction of Laggera forms into farming systems for medicinal and commercial purposes.

  A new genus Occultia (Scilloideae, Asparagaceaae), is described from Malawi and Mozambique. One of the species on which this genus is based, was originally described as Hyacinthus. The genus is clearly separated from Ledebouria and closely related genera in characters of the flowers and the fruits. In Occultia tepals are united for 1/3 to ½ their length, the filaments are very short and inserted at the top of the perianth tube. In the fruiting stage, the tepals of the new genus become membranous and detached from their basis, but may form a cap on the developing capsule. In Ledebouria the tepals are free or united just at the base, the filaments are of a certain length and inserted at the base of the tepals. If persisting at fruiting stage the tepals are attached to the base of the capsule, and may become fleshy with time. The new genus comprises two species O. hyacinthoides and O. fragrans.

  Cryptic species could represent a substantial fraction of biodiversity. However, inconsistent definitions and taxonomic treatment of cryptic species prevent informed estimates of their contribution to biodiversity and impede our understanding of their evolutionary and ecological significance. We propose a conceptual framework that recognizes cryptic species based on their low levels of phenotypic (morphological) disparity relative to their degree of genetic differentiation and divergence times as compared to non-cryptic species. We discuss how application of a more rigorous definition of cryptic species in taxonomic practice will lead to more accurate estimates of their prevalence in nature, better understanding of their distribution patterns on the tree of life, and increased abilities to resolve the processes underlying their evolution.

  Uapaca kirkiana Muell Arg. is one of the most popular indigenous fruit trees in the Miombo woodlands of southern Africa. An investigation into existing indigenous knowledge and socio- economic use of this fruit tree was conducted in Malawi. The survey revealed that U. kirkiana fruits contribute considerably to livelihood as a food supplement and for income generation among local communities. However, U. kirkiana trees are not widely cultivated due to lack of improved planting materials and difficulties in establishment outside their natural habitats. The results from the survey indicate that fruits are the main non-timber products that contribute to demand for domestication and the attributes preferred by the rural communities for improvement are mainly related to fruit taste and yield. Poor management and anthropogenic activities have resulted in the destruction of forest reserves and the local people do not have maximum stewardship of U. kirkiana trees in the forest reserve. The indigenous knowledge on U. kirkiana trees provides a valuable basis for domestication, propagation and improvement of fruit and tree attributes, and management issues on both co-managed and government forest reserve.

  Uapaca kirkiana Muell. angstrom rg is a dioecious fruit tree species for priority domestication in Southern Africa. It reaches reproductive maturity in eight to ten years with male plants making up 50% of breeding populations. Early identification of sex of seedlings is a prerequisite for selection and tree improvement. The amplified fragment length polymorphism (AFLP) technique was used to amplify DNA segments of U. kirkiana male and female plants to identify sex-specific markers. A total of 84 selective primer combinations were screened using bulked segregant analysis (BSA) for males and females. More than 110 polymorphic markers were obtained but each of the four primer pairs (E-ACT/M-CTG, E-ACA/M-CAA, E41+A/M-CTA and E-AGG/M-CTC) showed one band that was linked to sex. When the four primer pairs were tested in ten individuals from different populations only one primer pair (E-ACT/MCTG) amplified a 320 bp band in female plants only. It is possible that this marker is linked to a sex-determining locus. The results suggest that the gene that determines sex of U. kirkiana is autosomal in nature and this marker may be important during fruit domestication and tree improvement programmes. The marker has been sequenced and sequence characterised amplified region ( SCAR) marker will be developed and used for precise and rapid identification of female plants.

  Biodiversity is declining throughout southern African miombo woodlands due to poor land use practices that are detrimental to soil, vegetation and habitat. We aimed at examining tree and shrub species diversity and distribution in Uapaca kirkiana-dominated woodlands under three land tenure types; forest reserves, customary and leasehold land. The study was conducted at different forest sites within similar agroecological zone in southern Malawi. Study sites were located at a range of altitudes between 900 and 1,200 m a.s.l. Three circular plots each with 32.60 m radius were established at each study site to sample tree and shrub species composition, structure and distribution under three land tenure types. We found that forest reserves had higher species diversity with an average of 16 tree families, 27 genera and 34 species as compared to only 10 tree families, 6 genera and 6 species on customary forest lands. Comparisons of diameter at breast height size class distributions showed that customary land had significantly low numbers of small (5.0-10.0 cm) and very large (30 cm) diameter classes suggesting lower levels of regeneration and high rate of anthropogenic activities. The high species diversity and richness in forest reserves and leasehold land indicate high potential for protected lands to restore tree species diversity. It is concluded that levels of human activities as influenced by land tenure type reduce tree species diversity, composition and distribution at the different sites, and this confirms the hypothesis that open access lands are not compatible with conservation of tree and shrub species diversity because of high anthropogenic activities.

  Uapaca kirkiana is a priority fruit tree species for domestication in miombo woodlands of Southern Africa. Natural populations of U. kirkiana are declining through out the woodlands due to deforestation, forest fragmentation and wildfires. Knowledge of population structure and genetic diversity is prerequisite for development of conservation strategies. Amplified fragment length polymorphisms (AFLP) were used to assess the genetic diversity of eight populations from three geographical regions of Malawi. AFLP markers revealed moderate differentiation (GST = 0.079) among the populations collected from the three regions, however, there were no significant genetic variations among the regional collections. Analyses of molecular variance (AMOVA) found very high variation (92%) among individuals within populations and 6.8% among populations. The variations between populations indicate that populations can not be considered a single panmictic unit. Analyses of genetic similarity based on unweighted pair group method of arithmetic averages (UPGMA) suggested that the 8 populations fall into three clusters with 5 populations in one cluster, two in another and the lake shore population of Chesamu in its own cluster. Based on results presented it would be cost effective to sample a small number of populations represented by a large number of individuals for germplasm conservation purposes. In view of the wide distribution of U. kirkiana in the miombo woodlands in Southern Africa there is need for a more intensive genetic study to include populations growing in different countries to produce a wider picture of levels of distribution of genetic diversity of the species.

  Amplified fragment length polymorphism (AFLP) and random amplified polymorphic DNA ( RAPD) analyses were performed on six populations ( a total of 89 individuals) of Phytolacca dodecandra ( endod) collected in Ethiopia. The populations were selected based on our previous investigation to represent two altitude groups: lowland/central-highland ( 1600 - 2500 m) and highland ( 2501 - 3000 m). A total of 197 AFLP and 68 RAPD markers were scored from 5 primer pairs and 12 random primers, respectively. The overall patterns obtained for AFLPs and RAPDs from diversity, cluster and principal component analyses were very comparable. However, the moderate correlation ( r = 0.56) between AFLP and RAPD similarity matrices as well as the discrepancies in diversity estimates between the two techniques in some populations and in the lowland/central-highland plants could be due to differences in sensitivity of reaction conditions, bias in scoring of bands, number of markers used for analyses, and/or parts of the genome surveyed. For both AFLP and RAPD, the lowland/central-highland populations showed higher polymorphism and Shannon information measure ( H) than the highlands. Cluster and principal component analyses performed for both marker types have also clearly demonstrated the differentiation of all the lowland/central highland plants from those of the highlands, in agreement with our previous conclusion. Markers scored from any of the five AFLP primer pairs were sufficient to clearly distinguish the two altitude groups; with RAPD, selection of about 8 informative markers produced by seven random primers was needed for the same purpose.

  Norwegian universities have trained students and other scholars from the South within fields related to African plant diversity through the last decades. The activities were funded by NUFU, the Norwegian Council of Universities Committee for Development Research and Education, and 30 students successfully obtained PhD degrees in taxonomy and other biodiversity related fields, and all but a few have entered into scientific position at universities or other relevant research institutes in Africa. Most collaboration involved Zimbabwe, Malawi, Ethiopia, and Kenya, and though successful, they all faced the challenges of multi-institutional and multi-cultural teaching and research collaboration. Basic research within botanical diversity is better taken care of when the university councils own and administer the projects, compared to the alternative ownership by aid agencies.

  The delimitation of taxa in the Saxifraga rivularis complex is controversial. In the checklist for the Pan-Arctic Flora, the complex was tentatively divided into five species: the circumpolar S. hyperborea, the amphi-Atlantic S. rivularis and three amphi-Pacific taxa (S. bracteata, S. flexuosa and S. arctolitoralis). Two ploidal levels (2n = 26, 52) are known, indicating a possible history of reticulate evolution. In the present study, a combined approach using molecular (AFLP; 42 populations, 119 plants), ploidal (flow cytometry; 43 populations, 71 plants) and morphometric (81 populations, 216 plants) data were used to identify evolutionary lineages within the complex, and to delimit taxonomic species. Three distinct evolutionary lineages were identified in all analyses of the AFLP data: the Saxifraga bracteata lineage, the S. flexuosa/hyperborea lineage and the S. arctolitoralis/rivularis lineage. The three lineages had different DNA ratios: 0.20-0.21, 0.16-0.17 and 0.36-0.38, respectively. The DNA ratios in combination with the intermediate position of the S. arctolitoralis/rivularis lineage between the two others in multivariate analysis of both genetic and morphological data, suggest an allopolyploid origin of this lineage, with the other two lineages as progenitors. No single morphological character included in this study unambiguously separated among the lineages. They were, however, differentiated by combinations of characters, thus the lineages should be recognised at species level: Saxifraga bracteata, S. hyperborea (including S. flexuosa) and S. rivularis (including S. arctolitoralis). Within the Saxifraga flexuosa/hyperborea lineage two genetical groups were identified, correlating with geography rather than the morphological pattern giving rise to the current taxonomic treatment. The groups may reflect genetical differentiation in glacial refugia through the last glaciation, but are not yet reflected in morphological differentiation, and a division into two taxa is therefore not supported. In the S. arctolitoralis/rivularis lineage, the separation of a Beringian population representing S. arctolitoralis from the Atlantic populations in AFLP data and morphology and the very distant geographical range suggest a subspecific ranking of S. arctolitoralis. Most of the variation within the S. rivularis complex is found in North Pacific areas. This, in combination with the proposed connection to S. bracteata, suggests a Beringian origin for the S. arctolitoralis/rivularis lineage with a later expansion to Atlantic areas.
